One more "vote" for WPMU - yes it takes a little bit of tuning sometimes, but once that's done its far more comprehensive than other similar systems i've tested. And as was said above, you have a good support community and plugin framework going!
I suppose it depends on your goals though - feature potential vs simplicity etc...
